Abstract(s)
Objetivos: Caracterizar a relação entre a existência do polimorfismo do promotor do gene da metaloproteinase 8 e o desenvolvimento de periodontite, através de um estudo piloto realizado numa população portuguesa de utentes da Clínica Dentária Egas Moniz.
Materiais e Métodos: Selecionou-se uma amostra de 45 indivíduos, sendo que 21 eram pacientes periodontais e os restantes 24 tinham saúde periodontal. Procedeu-se à recolha de amostras com células da mucosa jugal, com recurso a um OmniSwab, para posterior extração do DNA respetivo. Efetuou-se a análise genética para o polimorfismo MMP-8-799. Esta análise foi realizada através das técnicas de PCR (Polymerase chain reaction) e RFLP (Restriction fragment lenght polymorphism), seguidas da análise eletroforética. Adicionalmente, ainda se procedeu à análise estatística, por meio do Software R Statistical 4.0 (R package based on χ2 test, Statistical Computing and Graphics, Inc.).
Resultados: Verificou-se que para o polimorfismo MMP-8-799 a presença do alelo mutado (T) foi significativamente superior no grupo doença (60%), comparativamente ao grupo controlo (31%). Neste sentido, existem evidências estatisticamente significativas para afirmar que há uma correlação entre as frequências alélicas e a ocorrência de periodontite (p = 0,013). Assim, o presente estudo piloto sustenta a hipótese de que os alelos T estão associados à ocorrência de periodontite.
Conclusão: Verificou-se que o polimorfismo MMP-8-799 está significativamente associado à periodontite.

Materials and Methods: A sample with 45 individuals was used, 21 of which were periodontal patients and the remaining 24 had periodontal health. Samples with cells from the jugal mucosa were collected, with the help of an OmniSwab, for later extraction of the respective DNA. Genetic analysis for the MMP-8-799 polymorphism was performed through PCR (Polymerase chain reaction) and RFLP (Restriction fragment length polymorphism) techniques, followed by electrophoretic analysis. Additionally, statistical analysis was carried out using the R Statistical 4.0 Software (R package based on χ2 test, Statistical Computing and Graphics, Inc.). Results: It was found that for the MMP-8-799 polymorphism the presence of the mutated allele (T) was significantly higher in the disease group (60%) compared to the control group (31%). In this sense, there is statistically significant evidence to affirm that there is a correlation between allele frequencies and the occurrence of periodontitis (p = 0.013). Thus, the present pilot study supports the hypothesis that T alleles are associated with the occurrence of periodontitis.
