Efeitos locais e sistémicos produzidos em diferentes modelos de indução de periodontite : estudo in vitro em ratos

| dc.description.abstract | Objetivo: Comparar os efeitos locais e sistémicos produzidos por dois modelos de indução de periodontite in vitro em ratos, pelo método de ligadura e injeção gengival de lipopolissacarídeo (LPS) de Porphyromonas gingivalis (P. Gingivalis). Materiais e métodos: Estudo experimental, efetuado numa amostra de 28 ratos fêmea Wistar. Aleatoriamente foram distribuídos em três grupos, o controlo (n=12), e os grupos experimentais com periodontite induzida por ligadura (n=5) durante 21 dias e outro com periodontite induzida por injeção de LPS de P. gingivalis (n= 11) durante 14 dias. O processo inflamatório no periodonto e órgãos reprodutores femininos foi avaliado por análise histológica. Os níveis séricos de hemograma, hormonas sexuais e parâmetros inflamatórios foram medidos. Todas as avaliações histológicas e sanguíneas foram realizadas por patologistas experientes que desconheciam o status da amostra. A analise estatística foi realizada com nível de significância p-value ≤ 0,05. Resultados: No geral, o grupo de periodontite induzida por ligadura apresentou contagens de linfócitos (82,62% ± 4,39) e IgG (93,32 ± 20,7) mais altas e leucócitos (2,18% ± 0,52) e neutrófilos (15,76% ± 4,68) mais baixas, do que os grupos de periodontite induzida por injeção de LPS e o controlo. O grupo de periodontite induzida por ligadura foi significativamente diferente dos outros grupos. A inflamação no útero, identificada na amostra pela técnica de ligadura, mostra uma possível relação com alterações no útero de ratos fêmeas Wistar com periodontite crónica. Conclusão: O grupo com periodontite induzida por ligadura, mostra que o método induziu maior destruição periodontal comparativamente com o protoloco de indução com LPS e tiveram um potencial impacto nas doenças inflamatórias sistémicas, tanto a nível sanguíneo como a nível histológico inflamatório. | pt_PT |
| dc.description.abstract | Objective: To compare the local and systemic effects produced by two models of in vitro periodontitis induction in rats, by the method of ligation and gingival injection of lipopolysaccharide (LPS) from Porphyromonas gingivalis (P. Gingivalis). Materials and methods: Experimental study, performed on a sample of 28 female Wistar rats. They were randomly distributed into three groups, the control (n=12), and the experimental groups with periodontitis induced by ligature (n=5) for 21 days and another with periodontitis induced by injection of P. gingivalis LPS (n=11) for 14 days. The inflammatory process in the periodontium and female reproductive organs was evaluated by histological analysis. Serum blood count levels, sex hormones and inflammatory parameters were measured. All histological and blood assessments were performed by experienced pathologists who were blinded to sample status. Statistical analysis was performed with a significance level of p-value ≤ 0.05. Results: Overall, the ligature-induced periodontitis group had higher lymphocyte (82.62% ± 4.39) and IgG (93.32 ± 20.7) and white blood cell (2.18% ± 0.52) and neutrophils (15.76% ± 4.68) lower than the LPS injection-induced periodontitis and control groups. The ligation-induced periodontitis group was significantly different from the other groups. Inflammation in the uterus, identified in the sample by the ligation technique, shows a possible relationship with alterations in the uterus of female Wistar rats with chronic periodontitis. Conclusion: The ligature-induced periodontitis group shows that the method induced greater periodontal destruction compared to the LPS induction protocol and had a potential impact on systemic inflammatory diseases, both at the blood level and at the inflammatory histological level. | pt_PT |
