Abstract(s)
Aspectos reprodutivos ligados a ambos os sexos condicionam a implementação de um
programa reprodutivo eficiente. Neste trabalho foram abordados dois tópicos relacionados
com reprodução em equinos. No primeiro estudo, objectivou-se avaliar a expressão de
proteínas da matriz extra-celular, a Decorina e o Cluster de Diferenciação 147 (CD147), em
cortes histológicos de endométrio equino saudável e com diferentes graus de fibrose
periglandular, através de imunohistoquímica e de western blot. Não obtivemos marcação com
o CD147. Em relação à Decorina, as marcações que nos pareciam indicar a sua presença no
endométrio, demonstraram-se inespecíficas. São necessários mais estudos, com outros
anticorpos, de forma a caracterizar a expressão destas proteínas no endométrio da égua. No
entanto, os nossos resultados sugerem a existência de uma relação do colagénio 1 e do α-SMA
à medida que aumenta a categoria de endométrio da égua.
No segundo estudo avaliou-se o efeito de diferentes procedimentos de congelação, na
qualidade do sémen de cavalo de raça Garrana. Quatro ejaculados de um garanhão foram
submetidos a dois diluidores de congelação (Gent® e BotuCrio®) e dois tempos de
centrifugação (10 e 20 minutos). Foi ainda estudado o efeito de um meio de centrifugação do
sémen (Centrifugation Cushion for Semen®). Cada ejaculado foi assim submetido a 8
tratamentos. Foi avaliada a mobilidade, concentração, vitalidade e morfologia dos
espermatozoides, no sémen antes da congelação, no congelado e no sobrenadante. Segundo os
resultados obtidos, a centrifugação durante 10 minutos e o uso de cushion parece ser a melhor
forma de processar o sémen para congelação, ao originar melhores taxas de vitalidade e
mobilidade.

Reproductive aspects linked to both sexes influence an efficient reproductive program. In this work two topics related to reproduction in horses were addressed. The first study aimed to evaluate the expression of extracellular matrix proteins, Decorine and Cluster of Differentiation 147 (CD147), in histological sections of healthy equine endometrium and with different degrees of periglandular fibrosis. Immunohistochemistry and western blot were used. No immunolabeling for CD 147 was detected in mare endometrium. In immunohistochemistry studies, decorine immunolabels, which seemed to us to demonstrate its presence in the mare’s endometrium were nonspecific. Further studies with other antibodies should be performed to characterize the expression of these proteins in the mare's endometrium. However, our results suggest a relationship between collagen 1 and α-SMA, as the Kenney endometrial category of the mare increases. The second study aimed to evaluate the effect of different freezing procedures on the quality of Garrana horse semen. Four ejaculates from one stallion were subjected to two commercial freezing extenders (Gent® and BotuCrio®) and two centrifugation times (10 and 20 minutes). The effect of a cushion media (Centrifugation Cushion for Semen®) was also studied. Thus, each semen sample was subjected to 8 different treatments. Mobility, concentration, viability and morphology of sperm were evaluated in semen before freezing, frozen semen and in supernatant. According to our results semen centrifugation for 10 minutes and the use of cushion seems to be the better way to process semen for cryopreservation, since it was the procedure with better viability and mobility rates.
